Magnetic solid-phase extraction of tetracyclines from milk using metal-organic framework MIL-101(Cr)-NH2 functionalised hydrophilic magnetic nanoparticles

Novel metal-organic framework MIL-101(Cr)-NH2 functionalised hydrophilic polydopamine-modified Fe3O4 magnetic nanoparticles (Fe3O4@PDA@MIL-101(Cr)-NH2) were synthesised and used as magnetic solid-phase extraction (MSPE) adsorbents for extracting tetracyclines (TCs) from milk samples. The integrated Fe3O4@PDA@MIL-101(Cr)-NH2 exhibited convenient magnetic separation and exceptional multi-target binding capabilities. Furthermore, the PDA coating significantly enhanced the hydrophilicity and extraction efficiency of the material, thereby facilitating the extraction of trace TCs. Various factors affecting MSPE, such as adsorbent dosage, extraction time, pH value, and desorption conditions, were optimised. The developed MSPE method coupled with high-performance liquid chromatography demonstrated good linearity (R2 ≥ 0.9989), acceptable accuracy (82.2%–106.1%), good repeatability (intra-day precision of 0.8%–4.7% and inter-day precision of 1.1%–4.5%), low limits of detection (2.18–6.25 μg L−1), and low limits of quantification (6.54–18.75 μg L−1) in TCs detection. The approach was successfully used for the quantification of trace TCs in real milk samples.

P-chromophore stability of Mehlich-1 and Mehlich-3 under Braga & Defelipo or Murphy & Riley dosing methods

ABSTRACT The performance of colorimetric methods for P quantification has been overlooked by researchers for decades. We investigated the performance of two blue colorimetric methods for P quantification, Braga and Defelipo (B&D) and Murphy and Riley (M&R), using two commonly soil P extractants, Mehlich-1 (M1) and Mehlich-3 (M3). Specifically, we evaluated the color development time and its stability in different soil extractant solution proportions (1:1 or 1:4 v/v), the optimum wavelength, limits of detection, and element quantification. Our results indicate that M3 leads to lower limits of detection and quantification for both colorimetric methods, particularly for B&D. For M1, the volumetric ratio (1:1 or 1:4) did not influence color development and both B&D and M&R methods showed a fast color development. However, B&D showed greater color stability (from 5 to 600 min) and an optimum wavelength of 711 nm, while M&R was stable from 27 to 600 min an optimum wavelength of 889 nm. For M3 soil extractant, there are important issues, such as M&R being unstable and B&D presenting slow color development. In addition, the spectral profile obtained from soil extractant was different from the one obtained by the calibration curve without soil for both M&R and B&D. Therefore, the adoption of original methods on P quantification in M3 extracting solution represents a potential source of error, leading to wrong P fertilizer recommendations. Thus, the best option for P quantification in M3 extracts seems to be the use of Inductively Coupled Plasma Atomic Emission Spectrometry (ICP-AES).

Comparative study of sample preparation procedures to determine the main compounds in ayahuasca beverages by QuEChERS and high-performance liquid chromatography analysis.

Ayahuasca is a psychoactive drink originally consumed by indigenous people of the Amazon. The lack of regulation of this drink leads to uncontrolled consumption, and it is often consumed in religious contexts. The aim of this work is to compare three miniaturised extraction techniques for extracting the main ayahuasca compounds from beverages. Three sample pretreatment techniques were evaluated (dispersive liquid-liquid microextraction [DLLME], microextraction by packed sorbent [MEPS] and QuEChERS [Quick, Easy, Cheap, Effective, Rugged and Safe]) for the simultaneous extraction of N,N-dimethyltryptamine (DMT), tetrahydroharmine (THH), harmine, harmaline, harmol and harmalol from ayahuasca beverage samples. Then, the most promising technique (QuEChERS) was chosen to pre-concentrate the analytes, subsequently detected by high-performance liquid chromatography coupled to a diode array detector (HPLC-DAD). The procedure was optimised, with the final conditions being 500 μL of extractor solvent, 85 mg of primary secondary amine (PSA) and 4s of vortexing. The analytical method was validated, showing to be linear between 0.16 and 10μg/mL for β-carbolines and between 0.016 and 1μg/mL for DMT, with coefficients of determination (R2) between 0.9968 and 0.9993. The limit of detection (LOD) and lower limit of quantification (LLOQ) were 0.16 μg/mL for all compounds, except for DMT (0.016 μg/mL) and extraction efficiencies varied between 60.2% and 88.0%. The analytical methodology proved to be accurate and precise, with good linearity, LODs and LLOQs. This method has been fully validated and successfully applied to ayahuasca beverage samples.

Sensitive and effective method with 96-well plate for determination of levamlodipine in human plasma using LC–MS/MS

we developed an effective protein precipitation method for determination of levamlodipine in human plasma using LC–MS/MS. Sample extraction was carried out by using liquid–liquid extraction in 96-well plate format. (S)-Amlodipine-d4 was used as internal standard (IS). The chromatographic separation was achieved using Philomen Chiral MX (2) column (3 μm, 2.1 × 100 mm). Mobile phase A was comprised of Acetonitrile (ACN), Mono ethanol amine (MEA) and Iso-Propyl alcohol (IPA) (1000:1:10, v/v/v), Mobile phase B was IPA-ACN (2:1, v/v). The flow rate was 0.4 mL/min. The total run time of each sample was 4.0 min with gradient elution. LC-MS/MS spectra were generated in positive ion mode, and multiple reaction monitoring (MRM) was used to detect the following transitions: m/z 409.20 → 238.15 for levamlodipine and 415.25 → 240.20 for (S)-Amlodipine-d4 (the IS). The method was linear from 50 to 10000 pg/mL（R2＝0.9988489）,and the lower limit of quantification (LLOQ) was 50 pg/mL. This method was applied to a bioequivalence study of levamlodipine.

Abstract PO3-18-05: Pharmacokinetics (PK) of lasofoxifene (LAS) monotherapy and combined with abemaciclib (Abema)

Abstract Introduction: LAS is a selective estrogen receptor modulator (SERM)/tissue-selective estrogen agonist/antagonist studied in women with osteoporosis and vulvar-vaginal atrophy, and most recently, metastatic breast cancer (mBC) with ESR1 mutations. LAS data in healthy postmenopausal women and young men show linear PK and no differences by age or race (Gardner. J Clin Pharmacol 2006;46:52-58; Prakash. Drug Metab Dispos 2008;26:1218-26; Data on file). Also, no PK differences were found between Caucasian and Japanese patients (Jhee. AACR-JCA 2022, abstract A83; Fountaine. J Clin Pharmacol 2006;46:693-99). Here we report select PK data of LAS (5 mg/day) from women with mBC in the ELAINE 1 and ELAINE 2 oncology trials to indirectly investigate possible drug-drug interactions between LAS and Abema. Methods: Patients with mBC and ESR1 mutations who progressed on endocrine therapy and CDK4/6 inhibitors received LAS 5 mg/day as monotherapy (ELAINE 1) or combined with Abema (provided by Eli Lilly and Co) 150 mg BID (ELAINE 2). Blood was collected prior to daily dosing at weeks 2, 4, 8, and every 4 weeks up to the final visit in ELAINE 1, and visits during the first 8 weeks in ELAINE 2, to determine the plasma trough steady-state concentrations (Css) of LAS, Abema, and two active Abema metabolites (M2 and M20). LAS was assayed using validated high-performance liquid chromatography/tandem mass spectrometry (LC/MS/MS; detection 0.025 to 25.0 ng/mL); levels below the lower limit of quantification (LLOQ) were reported as 0 ng/mL. Abema and its M2 and M20 metabolites were measured by validated Turbo Ion Spray LC/MS/MS (detection 1.00 to 500 ng/mL); values below the LLOQ were reported as < LLOQ ng/mL. Mean trough Css at each visit were calculated for LAS (ELAINE 1 and 2), and Abema and its M2 and M20 metabolites (ELAINE 2). LAS levels from ELAINE 1 and 2 were compared, while Abema, M2 and M20 plasma levels in ELAINE 2 were compared with previously reported levels. Such indirect comparisons may help determine if Abema altered the metabolism of LAS, or if LAS altered that of Abema, M2, or M20. The relationships of LAS and Abema levels with progression-free survival (PFS) were also explored. Results: Across all time points for subjects who had PK samples assessed, the LAS mean trough Css was 33.6 ng/mL (CV%, 39.8) in ELAINE 1 (n=40) and 30.9 ng/mL (CV 50.5%) in ELAINE 2 (n=29); in both studies, LAS concentrations were consistent across visits. LAS levels in ELAINE 2 were similar to those in ELAINE 1 with no evidence of accumulation. Mean (CV%) Abema plasma concentrations at weeks 2, 4, 6 and 8 were 199 (102%), 107 (130%), 82 (271%), and 94.3 (223%) ng/mL, respectively. Minimum and maximums of the mean trough Css for LAS, Abema, M2, and M20 are shown in Table 1. No relationships between LAS or Abema levels and antitumor activity measured by PFS were seen (data to be presented). Conclusion: The steady-state PK of 5 mg LAS in the ELAINE 1 and 2 studies suggest that the addition of abemaciclib to LAS did not result in an obvious or significant LAS-Abema, drug-drug interaction. Abema PK data from ELAINE 2 were consistent with published data (European Medicines Agency, Verzenio). These results support the continued development of LAS alone and combined with Abema for treating ER+/HER2-, ESR1-mutated mBC that progressed on endocrine therapy and CDK4/6 inhibitors. Table. Simultaneous quantification and confirmation of oxycodone and its metabolites in equine urine using ultra-high performance liquid chromatography-tandem mass spectrometry

Oxycodone, an opioid commonly used to treat pain in humans, has the potential to be abused in racehorses to enhance their performance. To understand the pharmacokinetics of oxycodone and its metabolites in horses, as well as to detect the illegal use of oxycodone in racehorses, a method for quantification and confirmation of oxycodone and its metabolites is needed. In this study, we developed and validated an ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method that can simultaneously quantify and confirm oxycodone and eight metabolites in equine urine. Samples were subjected to enzymatic hydrolysis and then liquid–liquid extraction using ethyl acetate. The analyte separation was achieved on a Hypersil Gold C18 sub-2 µm column and analytes were detected on a triple quadrupole mass spectrometer. The limit of detection (LOD) and lower limit of quantification (LLOQ) were 25–50 pg/mL and 100 pg/mL, respectively. Excellent linearity of the calibration curves was observed over a range of 100–10000 pg/mL for all nine analytes. Retention time, signal-to-noise ratio, and product ion ratios were utilized as confirmation criteria, with the limits of confirmation (LOC) ranging from 100 to 250 pg/mL. The data from a pilot pharmacokinetic (PK) study suggested that oxycodone metabolites have longer detection periods in equine urine compared to oxycodone itself; thus, the detection of metabolites in equine urine extends the ability to detect oxycodone exposure in racehorses.

Development of an electroanalytical methodology for the identification and quantification of cocaine in samples seized using a carbon paste electrode modified with multi-walled carbon nanotubes

Among the drugs of abuse, cocaine (COC) is the second most commonly used worldwide due to its stimulating and psychoactive effects. Currently, this drug is classified, according to the World Drug Report (WDR), as an illicit substance and is frequently seized by law enforcement agencies. Additionally, it can cause social problems due to its abusive use. This study describes the development of an electroanalytical methodology using square wave voltammetry (SWV) associated with a single electrode of carbon paste modified with 10 % MWCNTs (multi-walled carbon nanotubes) to quantify COC in seized samples containing up to 20 % cocaine (low purity index) in the state of Espírito Santo (Brazil). SWV parameters, including Es = 10 mV, a = 30 mV, and f = 15 Hz, were optimized utilizing a Britton-Robinson buffer solution of 0.1 mol/L (pH 10) as a supporting electrolyte through the implementation of experimental design (CCD). COC showed an oxidation peak at approximately 1.0 V, with low limits of quantification and detection (1.90 × 10−6 mol/L and 5.75 × 10−7 mol/L, respectively), in the linear range from 2.36 × 10−6 mol/L to 1.38 × 10−5 mol/L, with the proposed methodology. Thus, this research develops a fast and low-cost technique in favor of the police, allowing the R2 = 99 %. The results obtained in the intraday calibration curves present a relatively low standard deviation (RSD < 8 %), indicating a good repetition of the value production of an electrode for less than US$ 1.42 per analysis.

Lithium acetate mediated paper-based assay for absorbance analysis of E. coli concentrations

Paper-based assays have revolutionized POC diagnostics, offering significant values across diverse sectors due to their simplicity, cost-effectiveness, and portability. Detecting E. coli is critical for public health but faces challenges in traditional methods, prompting ongoing exploration of alternative approaches. To address the need for heightened sensitivity, this study introduces lithium acetate as a novel element in a paper-based E. coli detection assay, accompanying absorbance analysis using microplate readers. Utilization of lithium acetate as a lysis reagent and carrying out absorbance measurements in paper microtiter plates provide a significant stride in improving sensitivity and assay efficiency. The assay substantially reduced the analysis time to 4hours with a lower quantification limit of 103 cfu/ml for detecting E. coli in pure cultures. The results suggest that the assay could determine 10 cfu/ml of E. coli in a contaminated milk sample with 4hours of enrichment time. Thus, the study contributes towards improving E. coli detection with lower quantification limits and shorter analysis times. It is anticipated that the diagnostic methodology would circumvent the challenges in conventional E. coli detection methods, placing emphasis on absorbance analysis to achieve a streamlined, accurate, and rapid paper-based detection technique.

First electrochemical nanosensor for ultrasensitive quantification of MET inhibitor, capmatinib based on carbon nanofiber networks incorporated with hybrid nanofiller nanogold-loaded porous acetylene black

In the current study, the first voltammetric method is constructed for the ultrasensitive quantification of capmatinib (CMT), a targeted therapy drug falls within the category of tyrosine kinase inhibitors (TKIs), using a modified carbon nanofibers paste electrode (CNFs-PE). Nanogold-loaded porous acetylene black (Au@PAB) as a conductive hybrid nanofiller incorporating carbon nanofiber networks (CNFs) was synthesized. The surface morphology of the synthesized Au@PAB and the ternary nanocomposite Au@PAB/CNFs was elucidated using various analytical techniques. The results show Au@PAB hybrid nanofiller incorporated CNFs has a critical property that increases electrocatalytic performance. The impact of scan rate, pH, and supporting electrolyte on the electrocatalytic response of CMT was explored on the Au@PAB/CNFs-PE nanosensor. Under optimum adsorptive stripping square-wave voltammetric (AdS-SWV) conditions, the developed probe displayed good sensitivity, selectivity, and a remarkable wide linear dynamic range of 1.0 × 10-8 – 2.0 × 10-5 M of CMT. The low limits of detection (LOD) and quantification (LOQ) were 4.6 nM and 15.4 nM of CMT, respectively. The features of easy operation and good selectivity make this nanosensor a talented candidate for CMT detection in complex samples, as demonstrated by its application in spiked biological samples with good recoveries.

Amine Switchable Hydrophilic Solvent Vortex-Assisted Homogeneous Liquid-Liquid Microextraction and GC-MS for the Enrichment and Determination of 2, 6-DIPA Additive in Biodegradable Film.

2, 6-diisopropylaniline (2, 6-DIPA) is a crucial non-intentionally organic additive that allows the assessment of the production processes, formulation qualities, and performance variations in biodegradable mulching film. Moreover, its release into the environment may have certain effects on human health. Hence, this study developed simultaneous heating hydrolysis-extraction and amine switchable hydrophilic solvent vortex-assisted homogeneous liquid-liquid microextraction for the gas chromatography-mass spectrometry analysis of the 2, 6-DIPA additive and its corresponding isocyanates in poly(butylene adipate-co-terephthalate) (PBAT) biodegradable agricultural mulching films. The heating hydrolysis-extraction conditions and factors influencing the efficiency of homogeneous liquid-liquid microextraction, such as the type and volume of amine, homogeneous-phase and phase separation transition pH, and extraction time were investigated and optimized. The optimum heating hydrolysis-extraction conditions were found to be a H2SO4 concentration of 2.5 M, heating temperature of 87.8 °C, and hydrolysis-extraction time of 3.0 h. As a switchable hydrophilic solvent, dipropylamine does not require a dispersant. Vortex assistance is helpful to speed up the extraction. Under the optimum experimental conditions, this method exhibits a better linearity (0.0144~7.200 μg mL-1 with R = 0.9986), low limit of detection and quantification (0.0033 μg g-1 and 0.0103 μg g-1), high extraction recovery (92.5~105.4%), desirable intra- and inter-day precision (relative standard deviation less than 4.1% and 4.7%), and high enrichment factor (90.9). Finally, this method was successfully applied to detect the content of the additive 2, 6-DIPA in PBAT biodegradable agricultural mulching films, thus facilitating production process monitoring or safety assessments.

Validation of an enzyme-linked immunosorbent assay test-system for bevacizumab concentration determing in biological fluids

Introduction. One of the reasons for the development of many diseases primarily malignant is the increased expression of Vascular Endothelial Growth Factor (VEGF). Bevacizumab is a drug that neutralizes the biological activity of VEGF. The molecular structure of bevacizumab is a recombinant humanized antibody. Its use reduces vascularization in the foci of increased VEGF expression which slows down tumor growth and also helps restore vision in a number of ophthalmic diseases. To determine the concentration of bevacizumab in human biological fluids a test system based on Enzyme-linked Immunosorbent Assay (ELISA) is presented.Aim. Aim of this study is the validation of this test system.Materials and methods. Blank sera of volunteers, bevacizumab solution, enzyme-linked immunosorbent assay, solid-phase sandwich ELISA kit, microplate photometer.Results and discussion. The following characteristics of the test system were determined: the lower limit of quantification is 2.0 mcg/ml, the upper calibration range is up to 200 mcg/ml, the accuracy and precision within one series and between series does not exceed 20 %, and the total error of the method – 30 %, short-term stability for samples at room temperature – 6 hours, long-term stability – 14 days at –20 °C, the ability to freeze/thaw of samples is up to three times, the ability to determine samples with a concentration above the upper calibrator after diluting in 2 times.Conclusion. The results obtained fully comply with international acceptance criteria and allow the use of the ELISA test system manufactured by LLC "Probiotek" for use in the field of clinical laboratory diagnostics.  

Safety and Immunogenicity of Accelerated Heterologous Two-dose Ebola Vaccine Regimens in Adults With and Without HIV in Africa.

Shorter prophylactic vaccine schedules may offer more rapid protection against Ebola in resource-limited settings. This randomized, observer-blind, placebo-controlled, phase 2 trial conducted in five sub-Saharan African countries included people without HIV (PWOH, n = 249) and people living with HIV (PLWH, n = 250). Adult participants received one of two accelerated Ebola vaccine regimens (MVA-BN-Filo, Ad26.ZEBOV administered 14 days apart [n = 79] or Ad26.ZEBOV, MVA-BN-Filo administered 28 days apart [n = 322]) or saline/placebo (n = 98). The primary endpoints were safety (adverse events [AEs]) and immunogenicity (Ebola virus [EBOV] glycoprotein-specific binding antibody responses). Binding antibody responders were defined as participants with a > 2.5-fold increase from baseline or the lower limit of quantification if negative at baseline. The mean age was 33.4 years, 52% of participants were female, and among PLWH, the median (interquartile range) CD4+ cell count was 560.0 (418.0-752.0) cells/μL. AEs were generally mild/moderate with no vaccine-related serious AEs or remarkable safety profile differences by HIV status. At 21 days post-dose 2, EBOV glycoprotein-specific binding antibody response rates in vaccine recipients were 99% for the 14-day regimen (geometric mean concentrations [GMCs]: 5168 enzyme-linked immunosorbent assay units (EU)/mL in PWOH; 2509 EU/mL in PLWH), and 98% for the 28-day regimen (GMCs: 6037 EU/mL in PWOH; 2939 EU/mL in PLWH). At 12 months post-dose 2, GMCs in PWOH and PLWH were 635 and 514 EU/mL, respectively, for the 14-day regimen and 331 and 360 EU/mL, respectively, for the 28-day regimen. Accelerated 14- and 28-day Ebola vaccine regimens were safe and immunogenic in PWOH and PLWH in Africa. NCT02598388.

Development of heat-assisted in-situ solvent formation liquid phase microextraction followed by derivatization for the simultaneous extraction of aliphatic amines from wastewater samples prior to gas chromatography-mass spectrometry

Herein, a cost-effective, rapid, and selective liquid phase microextraction procedure has been developed for the extraction of some aliphatic amines from sewage samples. The method was based on in-situ formation of chloroform as an extraction solvent into the sample solution followed by derivatization by butylchloroformate prior to gas chromatography- mass spectrometry. Chloroform is formed by the reaction of chloral hydrate and sodium hydroxide. The extraction procedure was accelerated by heating. Also heating decreases chloroform solubility in aqueous solution. To improve the analytical signals, derivatization of the analytes is done by butylchloroformate. The effect of major parameters on the extraction efficiency were studied and optimized. Under the optimized conditions, enrichment factor and extraction recovery were in the ranges of 259–296 and 78–89 %, respectively. Low limits of detection (0.08–0.12 ng mL−1) and quantification (0.28–0.41 ng mL−1), wide linear ranges, and relative standard deviations less than 6 % for intra −and inter-day precisions were acceptable. Lastly, the mentioned method was successfully performed to determine aliphatic amines in wastewater samples.

Detection of syringic acid in food extracts using molecular imprinted polyacrylonitrile infused graphite electrode

A highly sensitive, cost-effective, and reproducible electrochemical sensor has been developed using acrylonitrile (AN) molecularly imprinted polymer (MIP) over graphite (MIP-AN@G electrode) for the first time to detect phenolic compound, syringic acid (SA), in food extracts. The primary objective of the recognition of specific health-beneficial compounds like SA is to assess the overall quality of fruits and vegetables. Introducing UV–visible (UV–vis) spectroscopy, Fourier transform infrared (FTIR) spectroscopy, and scanning electron microscopy (SEM), the characterization of the fabricated MIP-AN@G electrode material has been performed. To investigate the electrode’s analytical characteristics, a tri-electrode system employing differential pulse voltammetry (DPV) and cyclic voltammetry (CV) techniques was used. Rigorous analysis revealed that the electrode could exhibit high repeatability, high reproducibility, reasonably good stability, and a wide linearity window from 10 μM to 100 μM concentrations with a satisfactory lower limit of SA detection (LOD) of 0.32 μM, and a lower limit of quantification (LOQ) of 1.06 μM. The sensor’s detection competence has been further tested in cauliflower, oregano, and black olives samples, and high accuracies of over 99% were achieved in each case when compared to the high-performance liquid chromatography (HPLC) analysis. Further, the t-test statistical analysis yields satisfactory results for the SA concentrations present in the three real samples.

New LC-MS/MS reference data for estradiol show mini-puberty in both sexes and typical pre-pubertal and pubertal patterns.

Reliable estradiol (E2) reference intervals (RIs) are crucial in Pediatric Endocrinology. To develop a sensitive ultra-performance liquid chromatographic tandem mass spectrometry (UPLC-MS/MS) method for E2 in serum, to establish graphically represented RI percentiles and annual RIs for both sexes and to perform a systematic literature comparison. First, an UPLC-MS/MS method for E2 was developed. Second, graphically represented RI percentiles and annual RIs covering 0-18 years were computed (cohort of healthy children (1181 girls, 543 boys)). Subsequently, RIs were compared with published data by systematic searches. Lower limit of quantification was 11 pmol/L, indicating high sensitivity. E2 first peaked during mini-puberty in both sexes (girls up to 192 pmol/L; boys up to: 225 pmol/L). As could be expected, girls showed higher pubertal E2 (up to 638 pmol/L). However, boys' RIs (up to 259 pmol/L) overlapped considerably. We found four studies in the literature which also used LC-MS/MS to determine E2 and published RIs for the complete pediatric age range. RIs varied considerably. Pre-pubertal and pubertal phases were present in all studies. Higher E2 during the time of mini-puberty in both sexes was documented in three studies including ours. Variability of RIs for E2 between studies illustrates importance of laboratory-specific RIs despite using a liquid chromatographic tandem mass spectrometry (LC-MS/MS) reference method. In boys, the striking E2 peak during mini-puberty as well as high pubertal E2 without phenotypic estrogenization in regular male puberty indicate that the role of E2 in children, and especially in boys requires better functional understanding.

Magnetic iron-based metal organic framework as an adsorbent for magnetic dispersive solid-phase extraction of pyrene and phenanthrene in water samples

ABSTRACT A dispersive magnetic solid-phase extraction by means of core-shell iron-based metal-organic framework (Fe3O4- SiO2/EN@MIL-101(Fe)) nanospheres coupled with high-performance liquid chromatographic was developed for the determination of pyrene (Pyr) and phenanthrene (Phe) in water samples. Key parameters such as sorbent amount, sample volume, pH, extraction time, type and volume of elution, and desorption time were optimised by response surface methodology (RSM). Under the optimised conditions (sample volume: 5 mL, extraction time: 3 min, eluent: 3 mL acetonitrile: propanol, and desorption time: 3 min), the developed method showed a low detection and quantification limits (0.015 and 0.040 ng mL−1, respectively), good precision (< 9.8% RSD), wide linear range (0.06 - 10,000 ng mL−1, R2 > 0.99), high enrichment factors (49–57), good matrix effect (> 93.2%), high reusability (25 times), short extraction and desorption time (3 and 3 min, respectively), and high extraction recovery (96.5%). The established method was able to recover the Pyr and Phe from spiked real water samples with an extraction efficiency of 95.71% and 96.32%, respectively (RSD ≤ 9.52%).

Composite of a Stabilizer-Free Trimetallic Prussian Blue Analogue (PBA) and Polyaniline (PANI) on 3D Porous Nickel Foam for the Detection of Nitrofurantoin in Biological Fluids.

Herein, a facile and highly effective nonenzymatic electrochemical sensing system is designed for the detection of the antibacterial drug nitrofurantoin (NFT). This electrocatalyst is a combination of a trimetallic Prussian blue analogue and conductive polyaniline coated onto a three-dimensional porous nickel foam substrate. A comprehensive set of physicochemical analyses have verified the successful synthesis. The fabricated electrochemical sensor exhibits an impressively low limit of detection (0.096 nM) and quantification (0.338 nM, S/N = 3.3), coupled with a wide linear range spanning from 0.1 nM to 5 mM and a sensitivity of 13.9 μA nM-1 cm-2. This excellent performance is attributed to the collaborative effects of conducting properties of polyaniline (PANI) and the remarkable redox behavior of the Prussian blue analogue (PBA). When both are integrated into the nickel foam, they create a significantly enlarged surface area with numerous catalytic active sites, enhancing the sensor's efficiency. The sensor demonstrates a high degree of specificity for NFT, while effectively minimizing responses to potential interferences such as flutamide, ascorbic acid, glucose, dopamine, uric acid, and nitrophenol, even when present in 2-3-fold higher concentrations. Moreover, to validate its practical utility, the sensor underwent real sample analysis using synthetic urine, achieving outstanding recovery rates of 118 and 101%.

Specific and Simultaneous Detection of E. coli O157:H7 and Shiga-like Toxins Using a Label-Free Photonic Immunosensor

The current study outlines the advancement of an innovative technique for the simultaneous detection of E. coli O157:H7 and its Shiga-like toxins in food samples by utilizing a photonic label-free biosensor coupled with a microfluidic system. This detection method relies on ring resonator transduction that is functionalized with specific bioreceptors against O157:H7 on silicon nitride surfaces capable of binding specifically to the antigen bacterium and its verotoxins. This experiment included the characterization of selected monoclonal and polyclonal antibodies employed as detection probes through ELISA immunoassays exposed to target bacterial antigens. A thorough validation of photonic immunosensor detection was conducted on inoculated minced beef samples using reference standards for E. coli O157:H7 and its verotoxins (VTx1 and VTx2) and compared to gold-standard quantification. The lowest limit-of-detection values of 10 CFU/mL and 1 ppm were achieved for the detection of bacteria and its verotoxins. In this study, the lowest limit of quantification (LoQ) achieved for bacterial quantification was 100 CFU/mL, and, for verotoxins, it was 2 ppm. This study confirmed the effectiveness of a new quality control and food hygiene method, demonstrating the rapid and sensitive detection of E. coli O157:H7 and its verotoxins. This innovative approach has the potential to be applied in food production environments.

A micro-extraction/derivatization method for RP-HPLC-FLD analysis of amino acids in wild boar meat

Methods that are easy to use, spend small amounts of reagents, and have minimal human interference are precious tools in different areas. Sus scrofa is the wild animal most used in human diet, however, the knowledge about its protein fraction is still scarce. In this work, a method for 22 amino acids quantification was optimized and applied to the analysis of meat from animals of different ages and genders. The optimization of acid and basic hydrolyses reveal an ideal mass between 75 and 100 mg, with 3 mL of hydrolysis reagent, and 22 h for a maximum extraction of tryptophan. The hydrolysates were automatically derivatized using few µL, and the compounds were analyzed by reversed phase - high performance liquid chromatography with fluorescence detection. For comparison purposes, total protein was assessed, using the Kjeldahl method. The method showed high precision and accuracy, as well as very low limits of detection and quantification, with good potential to be applied to other animal meats. Significant differences were found between the amino acid profile of different animals (e.g. aspartic acid and taurine contents were slightly higher in juveniles than in adults).

HPLC-MS/MS method development and validation for the determination of tetradecapeptide in human plasma

Introduction. The number of peptide drugs being developed and registered has increased in recent years. Therefore, modern analytical approaches and methods are required to determine these substances in biological matrices during pharmacokinetic studies. Peptides are structurally intermediate between small molecules and biopolymers, making it difficult to develop methods for determining them using High Performance Liquid Chromatography with Tandem Mass Spectrometry (HPLC-MS/MS). Peptide derivatization can help achieve optimal chromatographic separation and increase method sensitivity.Aim. To develop and validate a method for the determination of the tetradecapeptide (TDP) threonyl-glutamyl-lysyl-lysyl-arginyl-arginyl-glutamayl-threonyl-valyl-glutamyl-arginyl-glutamyl-lysyl-glutamate in human plasma by HPLC-MS/MS.Materials and methods. The determination of TDP in human plasma was performed by HPLC-MS/MS. Sample preparation included a combination of blood plasma protein precipitation with propionic acid solution in methanol, liquid-liquid extraction with chloroform, and peptide derivatization with propionic anhydride. Internal standard (IS) was threonyl-glutamyl-lysyl-lysyl-arginyl-arginyl-glutamayl-threonyl-leucyl-glutamyl-arginyl-glutamyl-lysyl-glutamate. Chromatographic separation was performed in gradient mode, eluent A was 0.1 % formic acid solution in water, eluent B was 0.1 % formic acid in acetonitrile. Column: Waters XBridge C18, 4.6 × 50 mm, 5 µm. Ionization source was electrospray in positive mode. Multiple reaction monitoring (MRM) transitions for 4-substituted TDP propionate were: 681.30 → 73.95 m/z, 681.30 → 84.00 m/z, 681.30 → 101.90 m/z, 681.30 → 140.10 m/z, and for 4-substituted IS propionate: 686.00 → 74.10 m/z, 686.00 → 84.05 m/z, 686.00 → 102.00 m/z, 686.00 → 140.00 m/z.Results and discussion. Validation of the developed method was carried out in accordance with the requirements of Eurasian Economic Union and the following parameters were determined: selectivity, matrix effect, calibration curve, accuracy and precision, recovery, lower limit of quantification, sample carryover, stability.Conclusion. The method for the determination of TDP in human blood plasma by HPLC-MS/MS was developed and validated. The analytical range was 5.00–1000.00 ng/mL, allowing the method to be used to study TDP pharmacokinetics.
